How long would it take to communte from S. Charlotte (Providence & 485) to/from Uptown (S. Tryon & 3rd) during rush hours?

Also is the highway the best way to commute?

Personally I'd go Providence Rd the whole way and not get on I485. During rush hour you should expect at least 45-50 minutes though.

I am near Hwy 51 and Providence (a few miles in from Providence and 485).
When I did that commute it was 45-65 minutes.

You can get there just as fast on a bike, take a look at Colony Rd., there is a nice route to uptown. It will save you gas, and the $100/month to park
