I have researched options for routes to Charlottesville (Keswick) from St. Augustine and I would like the quickest route. Any ideas? Thanks

Umm....

I-95 North to I-288 just south of Richmond, VA to I-64 west, right to the Keswick exit. (288 begins at 95 and ends at 64)

Should take you about 11 hours if you don't stop too much. I'd try to leave the house early enough to miss the rush-hour around Jacksonville. If you do that, you should beat the evening rush-hour around Richmond too, and get there in time for dinner.

Where are you located? If you are near Orlando or Tampa/St. Pete you could check flights on Allegiant Air into Roanoke and then drive to Charlottesville. Depending on when you want to fly you might be able to fly for just as cheap as driving the whole way. Just a thought.
