Would a casino really attract any new tourists to Chicago that wouldn't have come otherwise?

I believe tourist would appreciate a casino before they consider the 606 as a reason to travel to Chicago.

I'd consider the high line an additional urban amenity that makes New York appealing as a place to visit. I'd imagine the 606 will do the same. It may not be the specific reason a visitor chooses to come to Chicago, but its one more urban amenity amongst a large list that makes Chicago a worthwhile place to visit. A casino, on the other hand, can be found nearby just about anywhere these days.

I'm not sure a casino would really add much new revenue to Chicago, more likely it will take tourist/conventioneer dollars that would have gone to bars/restaurants/attractions and keep them in the casino.

You asked my opinion.

The 606 is more of a local attraction. A casino would appeal to tourist more in my opinion.

Stuff like the 606 is more popular than one might think. The High Line is a huge tourist destination in NYC.
Portland is a big tourist destination now. It is impossible to find a place to stay in the summer.

Casinos on the other hand are dying. Vegas casino owners focus on their Macau properties for gambling. They have turned their Vegas properties into more of a night club destination. If Chicago made a new nightclub zone for big DJ and techno shows, taking into account its dance music heritage ... that would be pretty cool. I think the typical native american casino would be a major dud, though.

Probably not in and of itself, but I'm sure it would help push some people over the fence on whether to visit or not. The more things you have to do in your city, the more attractive to tourists it will be.
