Knoxville for now,...its metro is nearing the 1 million mark
But in the future it may be Clarksville Or Murfreesboro as these cities are growing fast and may soon surpass knoxville
No. Because it's not just based on population. Yes, Clarksville and Murfreesboro are booming...but Murfreesboro is still a suburb/exurb. It has a university, yes...but it is not a dominant city in the region. Clarksville is sort of solidifying itself as #5 (Tri Cities aren't "one city"...but as an area, it is still pretty important). But Clarksville is still close enough to Nashville to be overshadowed. They don't even have their own TV market.
Knoxville is clearly the #3 city in Tennessee, and will remain so for the foreseeable future. The only city that can challenge it is Chattanooga, not Clarksville or Murfreesboro.
For New York, its Buffalo. NYC and Albany are ahead in importance.
for New Jersey, its Newark, Trenton and Atlantic City.
Albany and Trenton are both the political centers of the state.
HM to New Brunswick for Jersey.
Rochester is way ahead of Albany; and from an economic standpoint....even Buffalo. 2nd largest economy in the state. Rochester, Buffalo, and even Syracuse have much more in the way of cultural influence than Albany. Albany is merely a puppet of the NYC region.

Lol sorry for super late response. Orlando draws in more tourist and has a busier airport which brings in more revenue overall. I prefer Tampa over Orlando but think that Orlando is more important to Florida's overall economy.
What would you consider the third most important city in your state? For CA it's easily San Diego, so what about other states?
Washington - Redmond.
Not a common answer (most people would say either Tacoma or Spokane behind Seattle and Bellevue), but Redmond is the headquarters of Microsoft, Nintendo, and an awfully large cottage industry. Being an epicentre of wealth is an overriding factor to the detail that Redmond itself is but a suburb of Bellevue.
Although San Antonio is bigger the obvious answer is Austin for Texas.

3. Austin- 4th largest city, Capital, Best on Education
4. San Antonio- 2nd Largest City, popular with tourists
5. Fort Worth- 5th largest city, great with arts and entertainment
El Paso, Lubbock, Amarillo, College Station, Tyler, Corpus Christi, Mc Allen, and others are all on their way up, but the big 5 for now is clear

Contrary to popular belief SA does not pull out the most revenue from tourism.
To be important you have to have things that are important, and ASs biggest importance is the Naval Bases and they are not tops in the state for that.
Houston has the biggest economy, a whopper of a port system, the second and third biggest airports in the state, they have NASA, the second biggest metro, one of only two tier one schools in a major city, the most f500 companies, the most foreign consulates, etc etc
DFW has the second biggest economy, the busiest airport, fed reserve, biggest metro, second most f500 companies, etc etc
Austin is the capital. I can stop there but, it is also has UT, it's economy is on par with San Antonio.
San Antonio has the third largest economy, which doesn't really outweigh the capital because Austin is right there behind.
San Antonio has the biggest concentration of tourists in a downtown area, but that is a miss congeniality prize. It does not match up to having monster economies, massive Ports, BEING THE CAPITAL, most highly ranked universities, federal agencies, etc.

So there is no justifiable reason to list SA ahead of Austin.
