If I had to guess, I would say everybody should visit these cities at least once in their lifetime:
New York City
Los Angeles
San Francisco
Chicago
Philadelphia
Miami
Las Vegas
New Orleans
Washington DC
San Diego
Nashville
Boston
Seattle
Honolulu
Atlanta
Any I am leaving out or forgetting? Any that should be taken from the list? Tell me your thoughts and make your own!

I would probably switch Atlanta with Houston but other then that pretty much agree.

I'd take Atlanta off the list. It's a great place to live, not really a "must visit." Same goes for Nashville. I'd also take San Diego off the list. Another great place to live and even visit, but in my opinion it's not a must-see city. I'd add Orlando -- yes, it might be kind of lame, but every American should go to Disney World at least once in their lifetime.
My list includes large cities and small cities, places to understand the diverse nature of the American culture as well as historical references and iconic monuments.
NYC
Chicago
Washington DC
Philadelphia
Boston
Charleston
Savannah
St Augustine
Miami
Key West
New Orleans
Detroit
St Louis
Santa Fe
Los Angeles
Las Vegas
San Francisco
Seattle
Honolulu
In addition to the cities listed above, I'd add these natural wonders, natural and manmade parks:
Grand Canyon
Yosemite
Blue Ridge Parkway
Great Lakes
The Everglades
Death Valley
Disney World
To round out the American experience, pick one of the following "new" cities to visit:
Houston
Dallas
Atlanta
Phoenix
Denver
2nd priority:
- New Orleans
- Boston or Philly
- Miami
- Las Vegas
- Honolulu
Landmarks and Attractions:
- Grand Canyon
- Disney World
- California and Florida beaches
- Some ski resort (either out west or Vermont)
I love Dallas, Houston, and Atlanta, but they aren't cities to visit. They got some great attractions, but they're more cities to settle down in rather than visit for a few days. I'm also not sure what so important in Nashville or Seattle that they needs to be visited. They're great cities, but to visit? I think they're on the level of Minneapolis and San Diego: great, but not quite there.

I think every American should make it to our nation's capital at least once. It put's a lot of things into perspective in many ways.
