Which of the three has the best job market and lowest cost of living?

It will depend on the type of work that you are looking for and I think the COL is similar for all 3.

Of those three cities, Burlington, VT is the nicest overall. IMO, Manchester, NH and Portland, ME really leave a lot to be desired compared to Burlington.

Portland, Maine is pretty good. Nice downtown, near scenic coastline and I think it's on a similar level as Burlington. Manchester is just a workaday old mill city, not much to do there.
