I wouldn't call DC a vacation destination although it obviously gets a good bit of tourists.
Not a destination like Miami is but it's certainly considered a place most Americans need to/would like to visit at least once. Perhaps if I should of said, tourist spots vs. vacation destinations.
And we all know it's not a bunch of folks on vacation occupying all those hotel rooms. Atlanta, Dallas, and Houston are in the top 10 and they aren't vacation destinations either. On the flip side, Miami isn't in the top 10 but it is absolutely a vacation destination.

I don't know if the word "nexus" between best explains it, but we can look at things such as flight patterns between each city and other forms of transit as a form of connection. I will try to look those up.

Job market: I think SF, DC and ATL are in one tier, the other three are in the tier below that.
Desirability: Depends on what you are looking for. I think SF and SD are objectively probably the most desired of the group. But, MIA and DC are highly desired in their own way, and I think on avg; ATL is more desired than PHX
Cities/Urbanism: 1. DC/SF roughly tie with certain advantages to each3. SD/MIA tie with varying advantages to each 3. ATL>PHX
Economy: I feel like that sorta goes along with job market
Quality of life: Subjective
Recreational activities: West
Universities/ Research institutions: Close because SF is so strong, but I'll say east as ATL>SD IMO, and MIA>PHX.
Culture/Population/Diversity: High culture is going to go to the east because I think DC slightly beats SF, and I think MIA/ATL probably beat SD/PHX, though I think SD punches above weight here
Local Politics: Unsure
Infrastructure/Transportation: Slight lean East
Airports etc: East
