Louisville looks larger on paper because that 721,594 figure is for all of Jefferson County. The city and county are consolidated. Hamilton County has 845,303 residents, it's more densely populated, and you have to figure in all the large suburbs. Cincinnati has been a big, significant city for a while longer than Louisville. There is no question Cincinnati is larger. Don't get me wrong, I love Louisville, but Louisville can't compare to Cincy's size, amenities, jobs, shopping, and overall more urban feel. Although I think both cities are great, I do prefer Louisville! I think Louisville feels much more liberal and open-minded as a whole. The Highlands is, in my opinion, one of the best urban neighborhoods around. Old Louisville is beautiful. Downtown is going through a resurgence. I think Louisville as a whole feels a bit younger in population than Cincinnati. Louisville is extremely underrated, and a lot cooler than most would think.

Im a huge fan of Louisville, but I like Cincy better due, mainly, to the foody scene there (availabilty of certain kinds of foods and the markets and stuff), and also the older neighborhoods and hills make the place more picturesque (thinking here of Mnt Adams, Mnt Auburn, OTR, Tusculum, etc). The downtown is much more interesting in Cincy, too.
I should say the restaurant scene in Louisville might actually be better than Cincy (which used to not be the case), but the markets/delicatessen/"localvore" thing is better in the Cincy area. Also, there is nothing like the local butchers they have in Cincy, like Avril Bleh or the ones in Findlay.
Louisville has a better park system, in that the parks seem to be more integrated into the city, especialy with the parkway system. Cincy does have Eden Park, but its somewhat isolated up on its hill...Louisville has more "Burnet Woods" style parks, incoroporated into the city and as centerpieces of city neighborhoods.
Louisville is maybe better for an alternative/hipster subculture, and for visual/gallery arts scene. An example is that the big fall festival in Louisville is an art fair.
Cincy is maybe more avant garde when it comes to the performing arts (I'm thinking mainly of the Fringe Festival, but there are things like a local choreographers festival in June)

By the numbers:
Annual Gross Metropolitan Product (total of the goods and services produced in a metro area) for 2008:
Cincinnati: Approaching $100M/year
Louisville: Approaching $ 60M/year
Metro area population estimate for 2009
Cincinnati: 2.2M
Louisville: 1.3M
...so we are talking about cities that are orders of magnitude apart. There is simply more of everything in Cincinnati, becuase the place is just bigger and has a bigger economy. The thing to note, and the counterintuitive thing, is that there is as much as there is in Louisville, given it's position as a relatively "weak market" mid-range metro area.

Cincinnati without a doubt. Cincinnati has a much better urban feel, a better downtown with a much more dense feel and far less parking lots which invade Louisville. More development going on in Cincy, and a much more diverse economy.
Louisville is nice, but Cincinnati is on another level.

Cincinnati is just on another level than Louisville.
But Louisville is certainly the place to be on the first Saturday in May!

They are both great cities but very different. It's kind of like asking: which is better apples or oranges. And it's not just the size. Louisville has a more southern feel with houses on individual lots - about 40' wide or more. There is a small area called Butchertown that is denser.
Cincy is much more northern in feel with many houses very close together. Love those hills. My conclusion - if you like a dense urban neighborhood go with Cincy, if you want a little more space but still have great eclectic shops/restaurants nearby it's Louisville.
