Best thing to happen to Chicago: The great fire of 1871 allowed the city to re-invent itself and gave architects a blank canvas to start toying with new designs. This event could also be in the running for worst thing because of the loss of life.

Worst thing: white flight. It will take generations to undo the mistakes that took place over about 20-30 years.

For Boston:

Best: Probably the landfilling of the 19th century, which allowed the creation of Back Bay and several other areas of the city. Here's a picture showing the difference: File:Oldandnewboston.jpg - Wikipedia, the free encyclopedia

Worst: "Urban Renewal" of the 1970s which did several things: destroyed the West End neighborhood, a vibrant, diverse area of the city. It created Government Center which replaced Scollay Square, another vibrant--though gritty & sleazy--neighborhood. Cut off the North End and waterfront for several decades by having a major highway cutting through the center of downtown.
