This happened in a Myrtle Beach Target recently and so it begs the question. Places like Target and Walmart used detectors to keep their merchandise from leaving the store. Shouldn't they also take a bit of their budget and screen for weapons entering their stores? Would doing so violate our second amendment?

A metal detector would cause a huge huge line of people waiting to enter the store. You would have to put your keys, cell phone, pocket change etcetera in a bin, walkthrough the scanners, get pat downs, have wands run over your entire body. Then there's the people with metal implants. They would be required to bring a doctors note explaining the implant. It would be like going to the airport each and every time just for a gallon of milk. God, I hope that never happens!

Yes it would. Any business that required me to pass through a metal detector would not get any of my money.

Reaction would be the same if the three year old killed everyone in the store and him/her self.
You seem to have no idea about metal detectors and security arrangements; the delays this would cause would have people going to other stores, or just start shopping online even more, let alone the huge costs to the companies to implement and maintain this.
I have no idea why you get in a frenzy over such rare, isolated incidents.

Florida permits concealed carry of handguns, and neither Target nor Wal-Mart have policies to prohibit them, so what would be the point of metal detectors at the entrance?

The person who left the gun there should be charged with a crime.

I read the article, but am not sure why you go from being alarmed that a loaded gun is left in the toy aisle(disturbing to be sure), to posing a question on metal detectors
As to your question about infringement of 2nd amendment rights, it really depends on whether it is legal to carry concealed or open in the state to begin with. Most law abiding people who own guns are responsible with them, so it seems strange to infer that someone would be willing to leave a loaded gun in the kids section on purpose. What could possibly be the reason for intentionally doing that
For that matter, what about the loss of the gun and it's value to the person?
Then again it could be that anti-gun activists are up to their old tricks despite how irresponsible it is. This has been witnessed on the left with the animal rights nuts releasing infected animals, environmental activists spiking trees to harm loggers, and even some pretending to be Tea Party members and the like by holding up racists signs in an effort to discredit the groups/organizations they oppose.
Whether by accident or on purpose, they should use security footage to determine who did it, and prosecute them to the full extent of the law. If it is a wacko doing it for political reasons, they should look into their background and publicly aannouncewhat group/s they are affiliated with.
