I-95 Northern Virginia as well as I-80 in Youngstown, Ohio list New York City as its destination. I-80 actually ends at I-95 west of the George Washington Bridge
Yeah, I believe it is at the point where it is exactly 400 miles to NYC on I-80 not far from the I-76 interchange. Think it is by far the longest I have seen. Almost all of my driving has been east of the Mississippi though with the exception of a one week visit to Los Angeles; most of the longest ones I can think of are still < 150 miles from the destination cities.
There's also one in Barstow, CA and Wilmington, NC along I-40 for 2,551 miles.
However, there are only one of these signs to emphasize the length of the highway in total, but not typical destination/control city signs, which is what I think is addressing.
I was a little surprised when I moved to Chicago that a lot of the signs just have arrows and say things like "Iowa", "Northwest Suburbs", "Western Suburbs", "Wisconsin", "Indiana".
In Savannah you see signs for I-95 pointing to Florence even though it is over 170 miles away.
Another one are the signs for the Chesapeake Bay-Bridge Tunnel. I know there's signs for that on I-95 in DE, and there's also signs down by Emporia in VA.

I was always use to having cities on the signs.
At least those are more specific than the "Other Desert Cities" sign along I-10 eastbound at CA-111 (near Palm Springs, CA)
Blythe or even Phoenix don't even warrant a mention

Well, if you're taking the turnpikes you don't even pass through Pennsylvania unless one sticks slavishly to the interrupted I-95.
