I'd think that for most all the cities that are their respective state's capital, that the
capitol building would be the landmark. For larger capital cities like Boston and Atlanta, that might not be the case. For Columbus, that might be a football stadium.
For Miami, I'd say that you have a few choices. My first choice would The Freedom Tower. It's one of the oldest buildings in the city and very iconic. Another choice might be Villa Vizcaya. Many other iconic choices actually don't fall within the city limits of Miami but rather in Coral Gables and Miami Beach.
For Raleigh, if you don't want to use capitol buildings, I'd either recommend the North Carolina Executive Mansion which is an excellent example of a Queen Anne style home of many gables, or I can recommend Dorton Arena, a mid-century architectural wonder designed by Polish Architect Nowicki with its hyperbolic paraboloid structure.

Buffalo is tricky as there is a lot of interesting stuff there, but nothing really dominates the landscape in a visually identifyin way where you go "oh, that's Buffalo" ..... Buffalo City Hall is kind of imposing though
Phoenix has Camelback Mountain
In Philly I would add City Hall and the Liberty Bell
Las Vegas has the strip or some identifyable neon signs
Indianapolis IN - If you are looking for civic pride and a gathering place, it is the Soldiers and Sailors Monument or simply "Monument Circle". If you are speaking of global identifiers, it is the Indianapolis Motor Speedway.

For Austin I would also add 6th street and San Antonio you also have the Tower Of Americas
No offense but Austin's identity can't be 6th Street. Most people would not recognize it from any other busy American downtown bar district. What about the river there with hot springs? Maybe the capitol? Maybe the giant Texas boot (is the State Fair in Austin)?
I think Minneapolis could possibly be changed to Minneapolis-St. Paul or have the two separate, but I would add the Cathedral of St. Paul for St. Paul and, as much as some might dislike this, the Mall of America for both.
Millennium Park/Cloud Gate, the John Hancock Tower, and Navy Pier should be added for Chicago.
The Milwaukee Art Museum could be added for Milwaukee.
