Actually, it's interesting that 1/2 of the cars sold in the US aren't actually made by the big 3 auto companies that are headquartered around Detroit. I'm not sure how many of those cars built by the big 3 are actually made in Detroit....
I'm talking about more historical importance. Detroit was the main city that started it all. I think currently there's only 8-12 models made near Detroit, that's just my estimate though.
Washington D.C.
New York City
Chicago
Los Angeles

My thoughts exactly. Seems like these four are a no-brainer, but picking the fifth one is tougher. I'd probably go with either San Jose/Bay Area (country's biggest tech region) or Boston (country's biggest education town) as No. 5.
I'm kinda stunned no body has mentioned New Orleans. A massive amount material and goods goes in and out of that port. Not to mention its importance as a Gulf of Mexico oil hub.

As much as I love Boston, I think I might have to go with the Bay Area.
The following five are the most important in many respects- cultural prominence, global influence, economic/ political/ social impacts to the country as a whole:
1) NYC
2) LA
3) Chicago
4) DC
5) San Francisco
If there was a 6th, it'll be between: Atlanta, Dallas, Boston, Miami & Houston in my opinion
If it's truly unbiased, would it really be an opinion?
opps! idk why i added 'opinion' on there, sorry!
