Anyways I'm looking to move from Upstate NY (BRRR) to somewhere with better weather. What areas in the VA/NC region could offer me:
-Good COL
-Quiet, safe living
-Not politically extreme either way (I'm gay so as long as the Fred Phelps type isn't the dominant demographic-somewhat conservative is fine)
-Decent job market for IT, while not being too large an area
-Suburban
-Not big on the club scene
-Preferably VA or NC but wouldn't mind looking into adjacent areas north such as PA or WV
I def think a good spot can be found given your criteria- just curious what you mean by not too large of an area though?
Off the top of my head- anywhere in the research triangle, Charlotte, & possibly Wilmington (albeit, weak on the IT stuff) or High Point area.
As for VA, I'd really look into Richmond.
The Fredericksburg, VA area might work, but it's starting to get a little expensive. You can also consider Winchester, VA but that area tends to skew more conservative, although I it's getting more and more DC transplants by the day.

Lancaster, Pennsylvania I think checks all your boxes.
It does get cold, but definitely less snow and less cold than Upstate New York. The trade off is the summer season in Lancaster is much more pleasant than VA or NC with less humidity.
It has a high QOL, a nice (but small) start up scene with a growing creative and artistic class and it is close enough to Philadelphia, DC and NYC and The Atlantic beaches to take advantage of great weekend trips.
Its downtown is also very vibrant and interesting with much to do.

Yeah Lancaster area seems pretty nice, although I've never actually been to Lancaster County, I do pass through the area on my way back from vacationing every year (York and Adams Counties) but wouldn't actually mind having a real hot and humid summer (anything is better than the "summers" Upstate.

I wasn’t going to say anything until the last sentence. If anything, the summers are nice, as they are hot enough in terms of being able to actually do outdoor activities.
Also, the winter actually has been relatively mind(has hit 50’s last couple of days).
Anyway, I’d say to look into an area like Knoxville TN as well.
I was in Carytown last year, can't say that I felt safe. Ideally I wouldn't want to be in city limits wherever I end up. I do concur with your thoughts on the Richmond area though
