They can change the character and culture of a city, for sure.
There are plenty of southern cities that have grown with influxes of outsiders, and still feel very southern. Nashville comes to mind.
I think in Nashville's case, the areas that continue to retain it's somewhat Southern feel would be the Tourist traps, some restaurants, etc., that's how that City makes money! At the city level, not so much...Nashville has changed and continues to change being a city full of implants from California, New York and the Midwest, especially the Chicago area not to mention immigrants.

Still doesn't change its region though. Cities change as they get bigger and more diverse regardless of what region the city is in. The influx of southerners, black and white, to northern cities didn't stop them from being northern.
Mississippi and Alabama. The southern accent still runs very strong in both of these states, and I feel as though these two are not as influenced by transplants, as say Florida and now even Georgia.
We will gladly give back the panhandle to Alabama just don't call the rest of us "Southerners".
Ha ! Well it depends on who I'm talking too. Black South Floridians claim there southern heritage and will argue with you tooth and nail that they are southern.

You're not accounting for the fact that many of the cultural characteristics that are associated with being Southern are largely rooted in rural/agrarian culture. That makes sense when you think about it; for most of this country's history, the South--the nation's largest region geographically--didn't have many big cities compared to the North which produced several large cities early on. Big cities began proliferating throughout the South along with advances in public health (which prevented the outbreak of epidemics like yellow fever) and technology (air conditioning) that made the region more livable in the 20th century.
Going forward, it's obvious that many of the vestiges of Southern culture will mostly be concentrated in smaller cities and towns throughout the region while the larger urban areas will continue to be hybrids of a diluted Southernness and Anywhere, USA.
