all I know is, no matter what I want to be near the Ocean, and for me Philly is the maximum distance I'd want to live from the beach. I also love my home of NJ and would hate to be far from my family.
1st tier: You've researched, and a STRONG POSSIBILITY.
2nd tier: You're researched, and A possibility
3rd tier: You're researched, and unlikely, but a REMOTE possibility that hasn't been written off.

For me...
1st Tier:
Las Vegas/Henderson, Nevada
Hononlulu, Hawaii (I'm sure unaffordable, but nontheless I like everything else)
Los Angeles, California (and area).
2nd Tier:
Ft Lauderdale, Florida
Clearwater, Florida
3rd Tier:
Boston, Massachusetts
Raleign, North Carolina
Charleston, South Carolina
Seattle, Washington
I'll only do U.S. cities:
1st Tier:
Philadelphia - Seriously considering moving to Philly in a few years to pursue a graduate degree at UPenn. I've always wanted to experience an older, urban, public transit-oriented city on the east coast and Philly is my top choice. Low cost of living, vibrant urban core, lots of history, what's not to like?
Washington DC - I plan on living in DC for a large part of my life as I hope to work for the federal government one day. It's not where I want to move right away, because I want to experience other places first but I felt I had to put it in this tier because, realistically, it's where I'll end up.
2nd Tier:
New York City - Would love to live in New York for at least a couple years but the cost of living is a turnoff. Still, it has some great grad schools that I am considering.
Boston - (see above)
Chicago - I feel like I would love living in Chicago for a couple years. The grad schools in Chicago don't really fit my criteria as nicely as I would like though...
3rd Tier:
Miami - Seems like an awesome party city and a fun place to be young. The humidity is a turnoff and there really aren't any schools in Miami that are on my radar.
Austin - (see above)
Denver - Like everything about it, but I should really go a little further east considering I've been on the west coast my whole life.
I'm mostly looking for places where I could pursue graduate school for music, arts administration and/or psychology, with some fun mixed in (mostly the 3rd group).
1st Tier - The West Coast "S" Group
San Diego
San Francisco
Seattle
2nd Tier - The Big City Group
New York City
Miami
Chicago
Houston
3rd Tier - The Foreign Group
Vancouver
Madrid/Barcelona
Sao Paulo/Rio
Puerto Vallarta/Baja California Sur
