LBJ, Nixon, the oil industry, anti-gun activists - although, how much pro-gun control sentiment was there in the US pre-November '63? That would be an interesting topic for a thread.
I think there are two questions here - not just who benefitted the most, but who at that time thought they would benefit the most? There are a lot of answers to the latter question. LBJ would be at the top of the list - the Kennedys did not like him or trust him at all, and he didn't have any love for either of them. He went from being a relatively powerless, figurehead of a Vice President to the most powerful man in the world in the space of 6 seconds.
Aside from him, the CIA, FBI, organized crime, and the military all had good reason to think they'd be better off with Kennedy dead. The mob got him elected by fixing Illinois, and he paid them back by appointing his brother Attorney General and turning him loose on them. Hoover hated him and feared what 6 more years of a Kennedy presidency would do to the cult of personality with which had surrounded himself, the CIA never forgave him for what they regarded as a betrayal at the Bay of Pigs and didn't trust him not to stab them in the back whenever he got another chance, and at the height of the Cold War there were a lot of military officers with stars on their shoulderboards who thought he was going to let the country be destroyed by the Soviets.
So - if the question was intended to spark a discussion on who may have had motive to pop him, the answer is, one hell of a lot of people who were very good at doing exactly that sort of thing.
Any historic figure can be lionized and turned into a champion for a cause after he's no longer around to set the record straight. JFK was a mediocre president financed by a family fortune and an obsessive father who was still around at the time of his passing. His election represented the final stage of the full assimilation of "white ethnics", many of whom deserted his party within two decades. His personal indiscretions, including an addiction to painkillers which would draw heavy censure today, were drastically downplayed by the culture of his day, while at the same time, the expanding power of the mass media was being harnessed to sell to an emerging youth market based upon charisma, rather than substance. The assassination notwithstanding, he would not likely have lived to be a respected elder statesman.
The strongest evidence in favor of a conspiracy points in the direction of Marcello, Trafficante, Roselli and Giancana. Most of the rest is wishful thinking on the part of the "usual suspects" over there in Left Field.

Everyone, of course - that's why they had to fight over the choicest position from which to assassinate him.
I still maintain that the likeliest assassins were Frank Sinatra's drummer, Woody Harrelson's dad, and the guy on the grassy knoll wearing the pickelhaube! (and anyone who says otherwise is clearly running interference for the coverup!)
