and now the fireplace won't light. It's ventless, we've only lived here for one winter and have never had a gas fireplace before.

Last year there was a giant tank from a propane company in the yard, which was removed in the spring.

We purchased 100 gal propane tank from Lowes recently, plus the necessary attachments (couple of salesmen at Lowes helped). Now we can't get it to light. Other than the chance that perhaps hubby did not set it up properly, ie maybe wrong regulator, etc. (I have tried to find professional to come out and look at it but no luck finding anyone), is there a chance that there is just a huge amount of air in the line, from disconnecting old tank and connecting new tank, and we just really need to hold the lighter by the pilot and hold the button down for a LONG time? Hubby has already tried quite a bit. Any suggestions appreciated - it's going to be cold this week.

Barring bad installation, you've probably answered your own question. Air in the supply line needs purging. You'd be surprised how long that takes sometimes.
