I am not familiar with Houston and want to plan a short trip from San Antonio. I doubt that I could do this all in a single day, but maybe two days. Here is basically what I would like to see (in this order):

1. Galveston
2. Reliant Stadium
3. Galleria and up/midtown area.

I would ideally go to Galveston first. Then drive back and stop at Reliant on the way. Ending at the Galleria, where I would stay the night closeby.

Is this do-able in a day or two? Thanks for any tips regarding traffic, days, times, etc.
(I'm mainly concerned with traffic.)

If you left early in the morning on the first day, it's definitely doable, but I personally wouldn't recommend anyone trying to squeeze Houston and Galveston into two days.

Is there any specific event you're planning to attend at Reliant, or are you just looking to see the stadium>?

If you don't wanna hit traffic don't leave Galveston between 4pm and 7pm. 610 infront of uptown has traffic just about everytime of day.

I don't plan to spend too long in Galveston. Just want to see the Reliant stadium.

I wouldn't even know how to leave Houston from uptown to avoid traffic. The whole idea seems impossible.

2 nights would be better. A lot of time will be on the road between places. You could spend a lot of time in Galveston alone on the Strand and driving by some of the old 19th century architecture that is still there.

Its doable, but it all depend on how early you can get up to start your trip.

Make sure you stop in Pearland when you go from Galveston to The Galleria. We have a Luby's now, yaknow!

I think you can do all this in two days.

I lived in San Antonio before moving to Houston and this is a few things I would recommend:
Chinatown - Find the intersection of Bellaire and Ranchester and you'll be right in the center of it.
Williams waterwall - Right next to the Galleria
The Woodlands waterway - Out of the way, but well worthy for anybody coming from San Antonio because of the huge amount of pine trees.
Hermann park - Nice big park and Houston's lightrail runs right next to it and also Reliant Stadium

It really depends on what time you'll be leaving. I-10 West can be a "breeze" at times, while it can be terrible at others. I would simply avoid leaving Houston during any rush hour period.
