I'll take 25 or Sound Ave from Aquebogue, all the way to Orient Point. Especially when you get to the border of Greenport (just west of it) on Sound Ave by the ocean side hotels.
Route 27 east of Patchogue to Southampton (better in winter though), route 51 from Eastport to Riverhead, and Nugent drive/Route24 (although a short stretch) from LIE to Riverhead, then again all the way past Flanders to Hampton Bays. If you like alot of pines, these are the drives.
One other very short drive that I've loved since I was a kid...South Country Road between Patchogue and Sayville, and any of the roads to the beaches and docks that are off of it.
Some beautiful old houses, a duck pond, the tiny "downtown" area of Bayport, etc. It's actually more of a good walk than a drive.

How about start that drive on S. Country Road in Bellport Village? That's another beautiful (and historic) area.
A nice drive with not too many cars on it is Ocean Parkway (not the one in brooklyn)
You can actually take it from by Kennedy Airport (Nassau Expy 878), going through Long Beach (Park Blvd) to the Loop Parkway/Meadowbrook Pky to Ocean Parkway.
It connects all of Robert Moses Big beaches starting with Jones Beach. Go all the way out to the Robert Moses Causeway and either loop around back or take the RM north to sthe Souther State to go home.
Look at that on a map, it's a nice drive.
