My wife and I are going to be visiting Milwaukee for one night to catch a concert on April the 17th and will be staying at the Iron Horse hotel. This is our first visit so I'm looking for things to do during the day that are fairly close to our hotel. Not really interested in visiting any breweries.

What are you interested in?

Do you like spirits such as gin and absinthe? You can visit Great Lakes Distillery near the hotel:

Great Lakes Distillery | Small batch hand crafted spirits made one drop at a time…

But if that's not your thing you can visit the Milwaukee Art Museum, the Haggerty Museum (on the Marquette campus), the Joan of Arc Chapel (on the Marquette campus), Marquette U, Milwaukee Public Market, Harley Davidson Museum, City Hall and other historic buildings such as the Pabst, the lakefront (weather permitting), the Riverwalk (weather permitting), the Frank Lloyd Wright homes on Burnham off of Layton Blvd, galleries in the Third Ward, the Cathedral, the Basilica, and so on. Lots of good restaurants to try out: some traditional places such as Karl Ratzsch or Maders (German), Three Brothers (Serbian), and many good pizza places (Zaffiro's, Transfer, etc.). Some more trendy restaurants include Odd Duck, The Noble, and Hinterland.

Very good ideas. I also recommend a stroll along Brady Street, you can see some interesting people there, check out a few funky stores and grab a coffee at one of the numerous coffee shops.

What do you like to do? The Harley museum is right there but you probably already knew that. Whatever you do obviously you should see the lakefront. Even if you don't pay full admission (if art isn't your thing) you can get some coffee inside the art musuem atrium and look out at the astounding view and admire the crazy cutting edge architecture. Other then that I recommend exploring some of the neighborhoods. Maybe pick an area for dinner in a nice neighborhood and then walk around and check it out.

The iron horse hotel is pretty awesome. You would enjoy dining outside if the weather is right. Long shot but I feel like even if it's 60 and outdoor dining is remotely possible we will make it happen.
