If I remember correctly, I didn't think it was opened. It was probably the same thing that was in the briefcase in Pulp Fiction.

You're not meant to know. He never opened the box because he needed it to hold on to his sanity. So long as it was undelivered, his character felt he had a good reason to escape the island.

What was in it is not important. The important bit was that it was an undelivered package.

Also, if you really want to know and have the DVD, there is an easter egg on the second disc of the DVD. In the "Video and Stills gallery" there is an audio clip that has director Robert Zemeckis revealing what was in the unopened package.

If you don't have the DVD see the spoiler below.
