Does the movie set up the theme that the werewolf can only be killed by someone he loves?

Not explicitly, but it is strongly suggested. In one of the scenes where David is trying to convince Alex that he's a monster, he gives an example from an old movie, then suggests that she may have to kill him. I don't remember the exact wording (been too long since I've seen it), but the meaning is pretty clear.
I thought that David's friend Jack had told him that killing a werewolf was nothing special, not even silver bullets were required.
Yes and no. He does tell him that, but he's also trying to convince David to kill himself --- throughout the movie. Which only adds further weight that when David finally meets his fate, it ought to have some gravitas. Will Alex kill him? Will he kill himself?
But neither happens. He runs down an alley, gets shot, and credits roll.
I felt sorry because David's character was sympathetic and because the wolf actually looked scared and confused all surrounded by traffic, city lights, noise and, eventually, trapped in an alley in front of so many men with guns pointing at him.
I felt sorry for David, yes, but not the monster. Had the story set up the monster to be more of a misunderstood victim (like the early Frankenstein films did --- or even King Kong), then that might have worked. But the werewolf kills indiscriminately, and yes, even though he hesitates a moment over Alex, he doesn't spare her. He's about to lunge in for the kill when the cops open fire.
