To me, some of the really funniest scenes are when the director is playing with the audience and the audience doesn't get it. I remember watching "Bowling for Columbine" in an art theatre in Ft Lauderdale, where the audience was so far left-wing that the seats on the right side of the auditorium were unused. Moore starts out with his schtick of how many weapons can be concealed on the body, and then proceeds to have a kid pull gun after gun out of his shirt and shorts, to the point that it was like an old Warner Bros. cartoon. After about the third gun, it was obvious that the kid wasn't any credible threat. Walking down a street with that much firepower, without rattling like a tinker and staggering under the load would have been impossible. I started chuckling, and by about the eighth gun mysteriously appearing I was laughing so hard tears were forming in my eyes. The rest of the audience was stone cold silent and shaken, and people near me were visibly trying to get away.
On a lighter note, the end of the Muppet Movie has a wonderful closing scene, where Animal screams out "GO HOME! GO HOME!" It was precisely what the audience (and theatres) needed to keep the little kids from crying for more or wanting to see the next showing.
Of course, some of the early classic cartoons have fantastic humor. Search around online for "What's Opera Doc" If you love or hate opera, and like Bugs Bunny, you'll be hysterical for 7 minutes. "Kill da wabbit, Kill da wabbit! Yoi oh toh ho, Yoi oh toh ho!"
Then there are the one-offs in the middle of otherwise middling movies. I think it was "The Man Who Knew Women" that had the lead being able to read all female minds. He comes across a little French Poodle jumping up excitedly on the person walking her, and the voiceover exclaims in a perfect French accent, "Monsieur, Monsieur! I have to P OOP!"
Funniest scene in a movie, though. Hmmm. "Monty Python and the Holy Grail" has a lot of good ones, and the "Thin Man" movies had some good bits, but I'd have to think a while before deciding on any scene as being funniest.

The Nutty Professor (remake) dinner scene...so over the top. Road Trip - the scene where E.L. makes a sperm donation for extra cash. Producers (original) - the scene with Max Bialystock and "Hold Me Touch Me"...my boyfriend and I love to play "Soldier and the Milkmaid", too!
One of the funniest scenes to me, that always gets me laughing thinking about it is on Ricky Bobby when he's being interviewed and he keeps putting his hands up and he's all "I don't know what to do with my hands" rofl.
Once, my son was up getting some award or something and my daughter said he looks like Ricky Bobby, he dosn't know what to do with his hands, my husband and I started busting up because it was true rofl. Anyways, that's a silly line that gets used alot around our house

The real comedy in that movie was the fact that he took 5 speeches and edited it to make it look like one.

The movie 'I Think I Love My Wife'. Where this poor guy takes a viagra...and his wife falls asleep before he gets the chance to go to bed. In fact...he took TWO. Enough said!
I saw "South Park Bigger Longer and Uncut" again over the long weekend and this really is the funniest movie from beginning to end I have ever seen. It was even nominated for an Oscar for Best Original Song.
The marionette sex scene had me laughing out louder than any scene in any movie I can remember.
The scene where Gary (the hero) has to act his way into the terrorist group where he explained how he heard the cries from his pet goat when the "infidels" bombed his village - AWESOME and almost tear jerking.
The scene where Gary gives an hilarious yet extrememly accurate political analysis of the three types of people in the world. It was perhaps the best description Ive ever heard of they types of people we have in this political world. Awesome.
