NJ Transit has a train station at the airport that runs right into Penn Station. Take the monorail from the terminal to the train station, and then take the train into NYC- it doesn't get any easier.

BobKovacs is the easiest and cheapest way to go....But if you are anything like my wife and pack 3 suitcases to the max weight (she gets an extra since i only need 1 small one) then your easiest option might be the express bus right outside the terminals

IMO, take the airport monorail to the train link stop, and hop on a NJ Transit train to NY Penn Station.

+1. I have done this on several occassions and much easier to get where you need to from Penn Station than from the bus terminal IMO.

I prefer the airport express bus myself. Last time I took it they had multiple stops including near grand central station. Seems like a hassle to have to lug luggage onto the monorail then onto the NJT again.

Some more info:
The bus stops at port authority (westside), bryant park (sorta central), and grand central(east side). These are all near or on 42nd street. Here's a link

The NJT train option can be a real pain. I take the NJT train every day and whenever it stops at the airport I see people lugging their bags onto the train. When it is crowded there is no where to place your bags. The bus is right outside the terminal and all you have to do is place ur bags in the luggage compartment of the bus and enjoy the ride. No need to transfer.

Easiest would be to probably hop in a cab but it certainly wouldn't be the cheapest. Like the others have said, taking the train is probably your best bet.
