I think most would agree Brielle's your best bet for raising kids, if you can afford it. I've lived here half my life (10 years). Nicest and quietest of the three, although generally the most expensive. Lots of younger couples with kids and, from what I've heard, an insanely nice elementary school (went to private school so I cannot speak firsthand on that). There's virtually no crime and the people are generally friendly. Also, it doesn't become infested with tourists during the summer because it's not on the beach.

Manasquan and Pt. Pleasant seem like fine choices as well, depending on the area. You're referring to Pt. Pleasant Boro, not Beach, correct? Beach is so touristy aside from the "library section" further inland, which is better suited for families, IMO. Boro is okay but not as nice as the other two. Do note that Manasquan High School (also the sending district for Brielle students) was ranked significantly better than Point Pleasant High School by NJ Monthly - #115 vs. #172 - in 2008.
