I'd say no and you will be shown, 1 bedrooms to turn into a 2. Flex 2br means it's a larger 1 bedroom, and you put up a wall to make a 2 bedroom. Now if you get over needing to be in a lux and go for an old school walk up you can get a 2 bedroom in that range.

None lux building can have laundry in the building just to let you know.
Be aware many gym memberships in lux buildings are an additional cost to your rent. Building I know vary from $500 to $1200 a year.
Maybe in on a low floor in an older (relatively, meaning 90s-early 00s) building in Downtown Brooklyn or LIC. But I think even that would be out of range. Flushing has some new buildings where you could swing that, though your neighbors will probably speak more Mandarin than English. And you could do it easily in Jersey City if you're willing to cross the Hudson. You might find an owner of a new-ish condo who will rent out for a bit cheaper than market. That's it for options in that range for "luxury".
If you are not concerned about neighborhood, you might find a 2BR in Midtown West or Yorkville, around $3500 or so, but not many apartments, and likely not new/newer buildings, but full-service with laundry and fitness. For new/newer construction in your preferred rent range, you're looking at Downtown Brooklyn, around Gold Street. LIC is also a good bet, but for Manhattan and LIC, you would be looking at a 2/1, whereas you might be able to get a 2/2 in Downtown Brooklyn.
I agree with the advice to also look for condo rentals as most condos have on-site laundry, and many have fitness rooms.

This! In my building now it's hard to do laundry bc there is only one washer dryer per floor for 36 floors and most people do multiple loads on multiple floors at a time. In my previous small elevator building we had 4 washers and 4 dryers and there were only about 16 units so it was never an issue.
Usually you have to pay a fee for all the amenaties. We have a gym but its $400 a year and a pool which is $250 for Memorial Day - Labor day + $25 per guest.
