If cost is not a concern either, I think Cary would win because of the lowest crime rate. With Cary, you are comparing a suburb to several much larger and more diverse cities. It is not an apples-to-apples comparison.
I have also assumed you would not count the distance to a major hospital either. I would count Duke as Cary's major hosiptal. although the one's in Cary can certainly handle most emergencies.
You could also chart this for yourself, using the City-Data crime rates, or other source. All the places have good hospital, but I am not sure what your criterion for "major" is.
BTW, the supply of new construction single-story homes in Cary is very limited. There were only 8 listings on Realtor. The lowest prices start at $340K, at the Courtyards on O'Kelly Chapel. Most others were in the $500K range.

Can you provide a little more info? Is it because you are looking for work in a hospital, or you are retiring and need to be near one? Do you want an urban environment, suburban, or more rural? Why have you picked this list of cities?
The more info you provide, the more people can address. None of these is "the best"--what matters is what's best for YOU.
Thanks for the comments so far! My concern is just to be near a hospital with major services such as heart specialty. We wouldn't want to travel a couple of hours to get to one.
Cities/suburbs and beaches/mountains does not matter.
I think taxes in both SC and NC are pretty much equal but the gas tax is much more in NC. We are paying $1.88 right now.

I think your question about crime rate needs to be refined a bit. The relevant crime rate is the neighborhood in which your home is located, not the overall city crime rate.
For example, If you were looking at Carolina Arbors in Durham County, you should look at a site which maps various neighborhoods. Pick the one surrounding Carolina Arbors and use that for comparisons. Durham has some bad neighborhoods, but you really don't need to be concerned about the ones 12 miles away if you never go there. Carolina Arbors is gated, and I would have absolutely no fear about my personal safety living there.
Similarly, Greensboro has some bad areas, but if you lived in the Epcon development, Villas at Sedgefield, you would be 20 miles away from the downtown. Of course, the trade-off might be that you are further from the good hospital.
