100 million in 100 days is probably way too ambitious. That's almost 1/3 of the population in 3 months. Probably not going to happen. Maybe in 200 days.
I think it might be doable. I did some quick and dirty math and based on my state's Covid vaccine dashboard, today they administered 1 shot per every 315 people in the state. Using that figure against the estimated 2019 population of the US, and assuming (always dangerous) that other states average out to the same rate per day, that would be 1,042,030 vaccinations per day. They're opening up the next phase of eligible people next Monday, so I'd imagine the daily rate will increase. While I think my state is quite well run, it's generally mid-pack in just about every metric where the states are ranked. A good bellweather for average.
I think the first 100 million will happen pretty quickly. The second 100 million might not happen at all because I predict that by late spring, the herd immunity effect of the holiday spikes coupled with vaccinations is really going to send new cases and positivity percentages down, and the better it gets, the less inclined fence sitters will be to take the shot.
As a middle-aged man with three comorbidities, I will say it would be nice to have some f-ing clue when I can get the vaccine. The State of Illinois has proven to be completely incompetent about this, just like pretty much everything. When can I get vaccinated? Nobody knows!! Governor Pritzger and the government of this state couldn't manage their way out of a paper bag.

In January the estimate was May or June before the vaccine would be available here in S.C. I got my second dose this past Tuesday. So yeah, Joe is underpromising and overdelivering. It’s good to have a real President again.
