Solas has private dining rooms and you might ask if you can get their entire second floor on a slow night.

Second the parking downtown. Free in the evenings. I like Moore Square Parking Deck, convenient to everything and free. Park there once and you'll see.

Dos Perros in Durham has a basement that seats between 50 and 70, depending on how tight you want it.

Revolution's downstairs room fits at least 40, but it will be more expensive for sure.

Tyler's in Durham has a private room for 40 I think.

Busy Bee definitely has a private room that could hold 40. Good food and they definitely have beer.

No beer, but Golden Corral on Glenwood in Raleigh has a private room. They require a minimum of 40 to reserve room. Good buffet. Plenty of parking.

Black Horse Run rents out their clubhouse this time of year. You would have to pay, they have no food (bring your own food and beer), but you would have total privacy and a big place for 40 people. They are up off of Creedmore/50 Hwy just north of 540.
