That's about a 20 mile drive, all highway. I would think you could make it in about 20 minutes non rush hour. I'd add 10-15 minutes for rush hour, to be safe. Traffic can back up around I-540/US-70 and the I-540/I-40 interchange - not always, but enough to make it safer to allot 35 minutes or so.
I drive 540 from Triangle Town to 15/501 every morning, leave my house at 7 and get to my destination around 7:35. So I think from Six Forks to 54, 15-20 minutes.
If you are exiting to 54 to go to UNC campus, you've only won half the battle.
During my time working at UNC, the traffic from the 54 exit into Chapel Hill has grown exponentially, largely due to Meadowmont development and more importantly the creation of the park and ride lots at the Friday Center.
Depending on time of day, it can take an additional 20 minutes to get into campus and 20-35 minutes to get from campus to I-40. Much depends on where you park, what bus you are catching etc.
