Are you asking if it is feasible to drive from St. Louis to Branson, do anything, and return in the same day? Then, no. You could drive down (not quite four hours), spend some time around Branson, stay overnight, and return to St. Louis in the afternoon. Better would be to drive to Branson in the afternoon/evening, stay overnight somewhere close (Springfield, Ozark), have a full day for activities, stay another night, then return to St. Louis the following day. It all really depends on what you want to do and see.

There are many different things that will appeal to some and not to others. Here are a few that you could look up to see if there is anything of interest:

Branson Railway Branson Scenic Railway: Branson's Most Classic Attraction
Dicks 5 and 10 - Dick’s 5 & 10 | Downtown Shopping | Branson, Missouri
Too many shows to list - Branson Shows & Attractions - Branson Tourism Center
Ride the Ducks - Ride The Ducks Branson Home - Ride The Ducks Branson
Shepherd of the Hills - The Shepherd of the Hills Outdoor Theater and Homesteadtheshepherdofthehills.com
Ruth and Paul Henning Conservation Area - Area Summary
Silver Dollar City - including Marvel Cave. This alone will eat an entire day - Silver Dollar City Attractions | Theme Park | Branson, Mo.
Branson Belle - Tickets
College of the Ozarks - https://www.tripadvisor.com/Attracti..._Missouri.html
Grand Village Shops (I always go to Pete Engler Woodcarvers, just to look) - Grand Village Shops, Branson MO

There are also the lakes, fishing, boating, scenic drives, hiking, and much I am not remembering at present.
