I have stomach issues - basically trapped gas - and flying 18 hrs to asia was a nightmare. Twice. Basically the notion of flying is a pain in the ass altogether. Hours spent just in the airport before you even take off. Another near hour after landing to be out of the airport... just a nightmare all around.

Our last 17 hr trip SFO- SIN
3 of us all took different routes.
Direct flight won on many accounts! (as they usually do)
stopovers + transfers,,,
The LONGEST route (time) but shortest (mileage) happened to become the one that did 2 stop-overs, and COULD NOT land at Narita (high winds / 3 attempts, not able to touch down). So they had to go to Haneda, then figure out how to get out of there to destination. (12+ hrs later).
Gotta go somewhere... try to go direct!
Get an aisle seat, as bladder control for 17 hrs becomes a real S-T-R-E-T-C-H. I was on a window when the aisle passenger loaded up on drugs and was OUT for 14 hrs straight (Ouch, that hurt.)
Far from perfect...
I did have (2) kids screaming and kicking my seat for 17 hrs a few months ago.
Their parents 'escaped' to another row and fell asleep.
I remember Singapore Airlines had a similar NY - Singapore service that they launched in 2004.
Yeah, it was launched as all business class flight and cancelled not long after due to low demand. I wonder if they're doing mix cabins this time around.
Assuming one doesn't have a physical problem or is isn't very large, it's mostly mental. I tell myself, "this isn't a hard day, all I have to do is sit, sleep and amuse myself."
I definitely take and time sleeping pills (valium works best for me) carefully. For the first 5 hours or so I stay awake and try to 'enjoy' the flight activities (food, drink, chat with neighbor, movie). Then as things wind down I pop a valium. 5 hours later I wake but continue being half-awake/half-out which is a state where time passes quickly. By time I start becoming fully alert again, there's usually only 2-3 hours left to grind out.
A couple trips to the toilet or just out of the seat help to stretch out the kinks.
The last thing to do is to stew, thinking of how long it is or how far I'm going, or how uncomfortable.
fun to read about it..... wouldn't actually want to fly it.
