Prague is stunning.
It's one of the prettiest cities in Europe.
It's crowded and vibrant and has about anything you might like: architecture (both stately and art deco/nouveau), art, classical music, cathedrals, a castle, museums, a statue of Good King Wenceslaus, clubs, good dining, the Charles Bridge. Again, there are many venues that celebrate the artist Alfons Mucha http://www.abcgallery.com/M/mucha/mucha.html.
If you like it, Prague probably has it.

Walk along the small streets, into the neighborhoods.
It's lovely and safe.
I use either airbnb.com or wimdu.com for cheap accommodations.
You can search for apartments with wifi on the right side of the screen.
If you prefer upper end try hotelscombined.com; hotels.com; laterooms.com; booking.com; eurobooking.com (similar but sometimes they have different prices)

You'll want to stay in town in Cesky Krumlov. There are a few bnbs (bedandbreakfast.com) or guesthouses around town. Google for photos of the town.
From there, you're about 15 km from Cesky Budvar, home of the real and original budweiser beer.

Easily option 1, I wouldn't deal with London right now due to the Olympics.

If your planning on taking the ICE be sure to always check the website, they always have good deals for discounts on fares.
