Recently picked up a 2016 Accord Sport. The car comes equipped with factory 19" wheel. Tire size is 235/40/19. I'm looking to swap the continentals with a slightly meatier size 45 tire. Will that throw anything off regarding the ride or transmission?
Assuming you are sticking with the same tread width you are adding a full centimeter to the distance from the rim to the ground. Used to be that would throw off your speedometer but with today's electronics Im not sure how much it would affect it.
It'll definitely soften the ride but I dont think it will have much impact to the transmission negatively.

I know with traditional geared transmissions going bigger would sometimes effect it. Not sure about cvt's.
Change in tire DIAMETER will change real odo readings. odo is set to factory tire diameter.
WIDER tire will provide larger tire footprint resulting in larger rolling resistance and reduced mpg.
Comfort is really more about sidewall stiffness and tire pressure.
Very wide and very short sidewall height tires are rather hard to control on the road.

Why not go to a 18-17 inch wheel with a talker sidewall. You'll keep the same overall height but you'll have a taller sidewall and better softer ride. I'm sure that there is a entry level smaller wheel size
Easy way to check if different tire size affects speedo, odo. If you have access to an interstate highway, look at odo when you are at a mile marker. Take note of the 1/10th. Drive 10 or more miles. If the number is not constant at each mile marker, your new different size tires are affecting your speedo, odo and mpg.
