If we are talking about most active nightlife, it's Las Vegas. You'll see more people awake and on the streets and in casinos there than in NYC. Despite what many think, Times Square isn't packed with people at all hours of the day.
Why would times square be busy after 1am? Night life is not there. How about going to the Meat Packing district sure you will see plenty of people at 4am.

Agree if people are going to times square for nightlife, or most anything for that matter they are likely tourists

I went with a local, stayed from thurs. am to sun. am. and it was in mid January. maybe the girl had no clue what she was doing,because these bars were literally dead. one of the bars we went to is always shown on that show jersey shore ('bed') and its PACKED, yet when we went there were probably 12 people in there. however, what it lacked in the 'club scene' (in my experience) it made up for with other nightlife options; for example, people were literally just walking the streets passing out tickets to a concert at the bud light hotel--we went and ended up having a blast.

NYC has a better balance of nightlife. From trashy to super exclusve /trendsetting/hip to high society. New York is all about sophistication while there is no taste in Las Vegas.
No city is as buzzing as New York.

very true unlike one strip in vegas all of Manhattan is pretty bumping at night with nightlife.

times square on a 10 degree cold night at 3 am is pretty filled with people.
