The wineries have such diverse architecture that it's interesting to visit all the major ones that are near NOTL. Inniskillen, Jackson Trigs, Chateau Des Charmes,Konzelmann, Hillebrand, that should keep you busy for half a day or so.
Don't forget there is also a toll on the I90 from the PA border to Buffalo if you went that way.

If you want a real special dinner go to the Skylon tower in Niagara Falls, it's awesome. I always have the surf and turf.
The whirlpool Jet is a pretty amazing ride. It goes up the river right past the whirlpool.
One thing that I have always done with guests when they come to Niagara is to take them to the Lock 3 museum and canal viewing. They are always amazed watching the huge ships being lifted up and down through the lock. I have toured the Panama canal and it's no where near as good as the lock 3 experience from a canal perspective. Panama is better of course from a wildlife and jungle point of view,LOL. The elevated viewing platform is free and if you want to go to the museum it's 5 or 10 dollars, not much anyway.
You should check out what's on at Art Park in Lewiston while you are here. There is sometimes some really good things going on there and while you are at it you can explore Lewiston NY which is a nice little town.
The bulk of the drive from Sarnia to Niagara Falls is on the 401. Yes, you take the 402 to the 401 to the 403 to the QEW as lucknow said.
I guess that's one of the problems with internet forums: dealing with folks who don't have a clue about what they're talking about.
It's roughly 300 km from Sarnia to NF --- only 50km of the distance is on the 401.
I truly hope the OP enjoys their trip to the Niagara region, but members who are blowing smoke about things they know very little about don't help the cause.
There are two little ferries between Detroit and Port Huron, at Algonac and Marine City. No delay at all, a boat every 20 minutes, they carry about ten cars, and no delay at all at customs. Fare probably about the same as the bridge toll.
