I wouldn't call it the biggest but Tucson's media is constantly comparing to, and claiming they are better than, Phoenix constantly. I worked Downtown for a few years and was constantly in public, the easiest small talk was always hating on Phoenix. When they proposed and built the street car from the University of Arizona to the Downtown bar areas, groups protested downtown street corners wearing brown shirts that said (ironically) "Keep Tucson S*****" and a small slogan campaign that said 'Don't Phoenix Tucson'.
Likewise, in Phoenix most have never been to Tucson other than a short visit and they almost never cared for it, but rarely discuss it.
No Boston vs. New York? That's really the only city vs. city rivalry I can think of off the top of my head that is very real in real life and not just in threads on City-Data.

PPL in my high school(in Phoenix) all called Tucson the Dirty T but yeah I agree. Tucson never really crosses my mind at all but yeah they do talk crap about phoenix a lot but that's because they wish they lived in Phoenix
Pittsburgh and Philly don't have a rivalry outside of hockey. One is king of eastern PA and the other is king of western PA and I've never seen a huge rivalry outside of that.

Yeah I wouldn't really call it an intrastate rivalry one sided if anything. Tucsonans rabidly hate Phoenix. Phoenicians forget Tucson is there.
As far as I know, there is no Pittsburgh vs Philadelphia rivalry (outside of hockey?). They are literally on opposite ends of a very large state 5-6hrs/300-350 miles apart. Philly is the dominate city in all of PA and Pittsburgh does it's own thing.
I actually think the same would ring true for LA/San Francisco and Cleveland/Cincinnati.
NOVA and the rest of Virginia, methinks. More regional, though.
