Manhattan south of 14th Street is basically the most urban/24 hour place in the Western world. So the answer is very obvious.
Also, you're conflating terms. "Downtown" in the sense of NYC means a series of neighborhoods on the southern third of Manhattan island. "Downtown" in the rest of the U.S. typically means the central business district.
I think downtown Chicago trumps most cities, but can it trump New York in anything? I know you're talking about a select section of Manhattan, but really? You should be a little more specific with downtown Chicago though, is the Loop included? Because the Loop's foot traffic falls off pretty sharply after the evening commute of about 4-5PM, and the Loop is pretty lacking in store fronts also, especially compared with State St. or Michigan Ave, or Ohio St. etc.
But most people consider Midtown as more urban/24 hour vice than Downtown so that is why I didnt choose Midtown

I know Chicago cant compete with Manhattan that is why I divided it up and just said downtown. I thought it would make the comparison a tad bit more equal

Outside of Times Square/Theater District area, and maybe Lincoln Center (which isn't really Midtown), there is no part of Midtown with comparable nightlife to Downtown.
Probably 80%+ of nightlife venues are south of 23rd Street. Pretty much any major street Downtown will have more nighttime activity than the equivalent street in Midtown.
