New Orleans definitely, its a unique blend of cultures and history you just won't find anywhere else.
NYC for lottsa reasons already mentioned. It would be tied for #1 with NOLA or a very close 2nd.
SF also for lottsa reasons already mentioned.

I don't think anyone's mentioned Salt Lake City. It's setting, history, and religious makeup clearly set it apart from others. It's not near the top of any lists but it's still pretty unique I think.
Also wanted to mention Atlantic City, it's not huge or anything, but it has America's 1st boardwalk.

Buffalo? I've only been there once but I was pretty young so I don't remember much. What do you think makes it unique? Not trying to be rude, I really wanna know cause I didn't even think of it. Maybe I'll go back.

Proximity to Niagara Falls, and being near the Canadian border! I really love cities on the Canadian border. For me they're unique because they are a gateway city to two cultures/countries.

I don't know if that part of Canada truly is another culture at all. They share a common culture.

How is Atlantic City unique but not Vegas? OR Miami for that matter?

Oops I didn't mean to imply that, I already listed my top 3 then I was just kinda thinking of lesser ones. I agree they're both way more unique then Atlantic City. But I'm surprised, no one else thinks Salt Lake City is unique? I'm not saying it's #1 material, but still it's pretty unique I think.
