My main advice is don't try to do everything in 2 weeks. When visiting Europe for the first time, it is tempting to say, Italy is so close let's just go there. All you'll end up doing is travelling on trains or planes and not actually seeing/experiencing the places and actually enjoy being in Europe .
Prague is one of the most beautiful cities in Europe in my opinion. You should of course check out Prague castle, maybe do the paddle boats on the Vltava River, also visit all the big sites including the old town square. The best part is to walk around and experience the city and slow down.
There is also a very cool bone church an hour or so from Prague in Kutna Hora. It will enable you to get away from the city to see a bit of the Czech countryside.
I have also heard the Ceske Krumlov is a great town to visit. I did not make it there, but have seen stunning pics of it. There are buses that leave from Prague to Ceske Krumlov every day.
