Downtown Tucson is super close to the mountains and desert.
Runyun Canyon is right there in the heart of LA--but not close to downtown, really.
In terms of small resort towns, Gatlinburg, TN has black bear sightings regularly. I was there twice in the same year and saw a huge black bear downtown, both visits.
You will see many types of animals spotted in Buffalo Bayou in downtown Houston. From rabbits to turtles to different fishes. Even including an alligator at times and hearing frogs croaking. Downtown DC, you will see squirrels all over the place but that's about it.

The animals that live in the Buffalo Bayou aren't that easy to see from shore and I don't think the locals are particularly friendly to them.
Lots of cities have a river running through it. I think OP is talking about terrestrial wildlife like moose or bears.

No but they are there. Are people any more friendly to alligators as they are to bears?
Downtown Lynchburg, Virginia is directly adjacent to vast natural areas like the James River, Percival's Island Natural Area, Blackwater Creek Natural Area and all connected to downtown by a series of trails. I am constantly seeing large shorebirds, turtles, snakes, several varieties of fish, rabbits, herons, groundhogs, and even even deer roaming in and around downtown, especially down by the riverfront.

Probably the only thing a friend of mine in LA that follows me on Instagram is jealous of is how accessible nature and serenity is from downtown where I live by foot within 10 minutes. He was saying from where he lives it would take an hour or more of driving to reach that level of nature and seclusion without any people around. It is admittedly probably the nicest thing about this city.

I don't doubt that alligators have been found in the Buffalo Bayou, but that's very uncommon. I have not seen one and I used to go at least twice a month. They tend to prefer soil banks not concrete so the downtown area isn't ideal habitat. Turtles are also very uncommon. Most of the time the only wildlife you'll see are ducks.
