This may be a stupid question, so I apologize in advance.
Is the Winston-Salem or Greensboro area transit friendly? In other words, would a family of 3 need a car to purchase household necessities like food, etc.? Do you all have a Super Target or Walmart?
Both cities have bus systems that cover most of the area within the city limits. You could get by without a car, but it would severely limit your possibilities. I would not consider either to be "transit-friendly" unless you live downtown with a very narrow focus in your lifestyle.
And yes, there are super Walmarts and Targets in both cities.

It's not a stupid question, but the answer will disappoint you, because mass transit in most NC cities is NOTHING Like in larger Northeastern cities, or DC, Atlanta, etc. As is said above, I think not having a car would severely limit your comings and goings. You'd at least want good bicycles.
Several Super-Walmarts in Winston Salem and Greensboro, but like most are not centrally located, thus not really great bus routes. Ardmore would really be the only "walkable" neighborhood, with a couple of grocery stores. As to the Targets, most in the area have added small grocery store sections, nothing like the Super Targets/Walmarts, but they have a little bit of everything.
I live in Gibsonville 2 miles from the Alamance Co line. Alamance County has county wide transportation even into very rule areas. Contact Alamance County Transportation and live in the area and it will take you to school, work, sores, appt, anything you need.... its a great service... something Guilford should have not stopped as Alamance is making money off of their service. It is only $7 one way and you can get a week of transpo for a reduced fee. Good Luck!
