Researchers in The Netherlands and Germany have warned that Pfizer-BioNTech’s coronavirus disease 2019 (COVID-19) vaccine induces complex reprogramming of innate immune responses that should be considered in the development and use of mRNA-based vaccines.

Jorge Domínguez-Andrés and colleagues say that while the vaccine has been shown to be up to 95% effective in preventing infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and subsequent COVID-19, little is known about the broad effects the vaccine may have on the innate and adaptive immune responses.

In the current study (not peer-reviewed*), the research team from Radboud University Medical Center and Erasmus MC in the Netherlands, and the Helmholtz-Centre for Infection Research (HZI), Hannover Medical School (MHH), and the University of Bonn, in Germany, confirmed the efficacy of BNT162b2 vaccination at inducing effective humoral and cellular immunity against several SARS-CoV-2 variants.

However, they also showed that the vaccine altered the production of inflammatory cytokines by innate immune cells following stimulation with both specific (SARS-CoV-2) and non-specific (viral, fungal and bacterial) stimuli.

Following vaccination, innate immune cells had a reduced response to toll-like receptor 4 (TLR4), TLR7 and TLR8 – all ligands that play an important role in the immune response to viral infection.

Neta and colleagues also found that cytokine responses to fungi were increased following vaccination.

I wish one of the medical people could take this and translate it to English or in my case, speak to us like we are 5. I read this and get lost in the medical jargon. I read these studies and always feel like I know no more when I am finished than when I started except it appears there are negative effects here.

The only person "warning" anyone is the author of that article. Its an unfortunate choice of words because the original paper contains no warnings.

The question is, do these responses return to baseline X number of days after vaccination? Probably.

The old vaccines induce antigen-specific memory in our adaptive immune cells that enable protection against a target pathogen, like the flu.

The authors of the study are warning that mRNA vaccines, or at least this particular one, go beyond that and induces complex reprogramming of our innate immune responses. The effect of mRNA vaccines could interfere with immune responses to other vaccinations. They also said that the combination of strong adaptive immune responses and the reprogramming of innate responses could either contribute to a more balanced response to SARS2 or a weakened innate immune response to it. They warn that all of this must be taken into account and studied.

All of these issues exist with other vaccines too, and they certainly have been and will be taken into account and studied.

they don't warn anyone about anything, they merely mention some unpublished findings and suggest further study.

I don't understand how that could be, the mRNA vaccines only cause the cell to make the spike protein, they don't go anywhere near where our innate immune system is programmed...

The study is not peer reviewed nor published and so it is hard to comment on it.

We have known about the innate system since the 1940's when viral interference was first noted mainly that when a person when infected with one virus can prevent another virus from taking hold and replicating. The host immune response was nonspecific to include other viral infections. They isolated a chemical that they called interferon which is a cytokine that is released from cells as a result of infection and it is a translational inhibitory protein. That is how it works by preventing the virus through preventing the translation of the viral replication. It is nonspecific and works on all viruses. It comprises a major component of the innate system against viruses.

In 2012 a study was published that stated that the influenza vaccine caused a person to become more susceptible to other respiratory infections such as coronaviruses. They postulated that the innate immune system was impacted by the vaccine rendering them more susceptible to infections. It was postulated that such effects would only last weeks. The only problem was that that study findings was never replicated again with further studies. Many antivaxxers cite that study and implicate the same impact by other vaccines.

With regards to Candida, the host immune response to fungi is primarily an adaptive immune response based on cellular immunity. Those lacking cellular immunity are more susceptible to fungal infections such as with AIDS.

The COVID vaccine is said to generate both a humeral and cellular response against COVID. The allegation is that the innate immune response causes a lowering of interferon unlike infections and vaccines of the past. It also claims a higher cellular immune response based on Candida antigens.

Any immune response will cause mediators of inflammation which are important for the innate immune response and adaptive immune response to react thus altered levels either going down or going up. Most of the time it is time limited. For example it has been known for many years that when a person received a blood transfusion their immune system were lowered for unknown reasons. They were more susceptible to infections. This was noted and so immune tolerance was taken a step further by thus transfusing patients who were scheduled to receive kidney transplants of giving them a blood transfusion to reduce the chances of rejection.
It was also taken a step further about 10 years ago by filtering all blood products to get rid of the white cells. A lot of the immune modulation was being caused by the white cells present in units of blood. Studies showed a reduction of infections in post surgical patients. The point being that alterations in immunity happens and not just with vaccines.

With regards to autoimmune disease, cytokine panels and cellular marker panels are different compared to those of typical infections. There's different types of hypersensitivity that can result in different types of reactions. The Classic autoimmune diseases that we all know about generally involve a grace period what is called the honey moon phase of type 1 diabetes. There's an antibody phase that can last for a long time before target organ damage results in generating symptoms. With regards to pernicious anemia it may take years before B12 is all used up and render symptoms.

I do not take reports seriously until it is peer reviewed, published and reproduced and then one can talk about it and what it means. Over zealous researchers will always elevate the research implications. They love their work and want to keep funding their work.
