Why not just drive? At that hour of the night the roads are pretty good, you will shoot right through....
Taking the train causes issues, esp bc Hempstead is not a main hub and in order to get there from either the PJ line or Ronkonkoma line you will need to transfer in Jamaica (or maybe Hicksville??) and backtrack to Hempstead station...
It is not really convenient.....
You can't get there from here describes trying to go directly from Hicksville to Hempstead via the LIRR. The only way to get there from any point east is to transfer at Jamaica or Floral Park and backtrack.
To get from Mt. Sinai, I would say the best way is to take the Port Jefferson Branch to Floral Park, and then take a train to Hempstead. This will cost $6.75.
It is also possible to go to Mineola and then take the N15 or the N40/N41 to Hempstead, but that will require an additional $2.25.
NOTE: The last N15 leaves Mineola at 12:30AM, and the last N40 leaves Mineola at 12:00AM, so if you arrive at Mineola later than that, you'll be better off going to Floral Park and taking the LIRR back.
If you take the Ronkokoma train you can get off at Mineola... If this was during the day it would be no problem to get off at Mineola and take a bus to Hempstead... But at your hour there are no buses... You can get a taxi no problem or you can walk to the Garden City or County Life Press LIRR and then take the train to one or two stops to Hempstead...
If coming from Port Jefferson take the train to Merillon Ave... and walk down Nassau Blvd to the Nassau Blvd LIRR... Or New Hyde Park and walk down NHP rd to Stewart Manor LIRR... The Port Jeff stops are closer and pretty easy to walk to... But you are going at very odd hours, which means you should try to plan out the times so you can make the connections...
