Daisy Cakes is awesome. They also have a brick-and-mortar location now -- 401 Foster St in Durham. No need to chase the truck unless you like the fun of the pursuit. :-)

My family's favorite place for cupcakes is Whole Foods in CH. Seriously, would never think of them as grocery store cupcakes, and they do great vegan ones for my dairy-sensitive DH.

Hands down, THE BEST cupcakes and cakes, period come from Angela King at King's Cakery.
Everything she makes is SO YUMMY and beautiful too!
She can customize anything you like and her artistic talent is unbelievable.

I was also just recently researching this (but for the mini-cupcakes) and decided just to get something from Whole Foods, their mini-cupcakes look decent. I actually make good cupcakes myself, but the school doesn't allow homemade. You might want to check the reviews on Yelp. Gigis had lots of negatives there too.
