MLS will tell you that Man City vs Redbulls Franchise is huge. Traditionally the Portland Seattle rivalry is pretty big. There is none that is a "must watch" for me though I do try to watch as much as possible, at least with extended highlights.
Biggest is Portland vs. Seattle- must watch
then maybe
Dallas vs. Houston
Up and coming are
NYC Red bulls vs. NYFC- may become a must watch
LAFC vs. LA
Miami vs. Orlando- might become a must watch
Vancouver vs. Montreal or Seattle
Toronto vs. Washington D.C or any Canadian team.
Kansas City vs. Houston- this is a rivalry that is deep although not a must watch.
Right now the top rivalries, not necessarily in order, are:
LA Galaxy - San Jose Earthquakes
Real Salt Lake - Colorado Rapids
Seattle Sounders - Portland Timbers
FC Dallas - Houston Dynamo
As the league matures and new teams join in new rivalries will emerge.
As a die-hard Real Salt Lake fan my biggest rival and foe are the LA Galaxy.
