Geneva is traditionally the go to answer, and arguably the correct one too.
But for some cities roughly the same size or smaller that fall under this list.
Malacca- ~900,000 people
George Town/Penang- ~700,000/1,700,000 people
Djibouti City- ~600,000 people
Manama, Bahrain- ~300,000+ people
Georgetown, Guyana- ~100,000 people
Malacca and George Town/Penang- Historically diverse and still small places in critical trade chokehold. Also got heavier European influence than other parts of Malaysia and remain relevant in trade/culture today.
Djibouti as more and more bases get's setup and it's relatively the only peaceful country in the area with Sudan under dictatorship, Eritrea/Ethiopia at war with Tigray, Yemen in Civil War and Somalia in Civil War it's perfect for a Red Sea port and will likely get investment from all sorts of sources including places like Turkey, Saudi Arabia or Iran in the future not to mention France, American, Japan and China all already there. Then the actual population is diverse being 50% Somali 16% Afar (people who's homeland is mostly in Ethiopia) and refugees as well as French citizens.
Manama- One of the smallest of the hyper diverse ran by Oil middle eastern capitals. It's also historically important at the crossroads of Arab and Persian culture, and not far from South Asia by boat, making it an important trading point. I can only see it's importance grow from here.
Guyana's Georgetown might become the unanimous answer. It's already in a hyper-diverse country, which just struck Oil. This will cause some sort of migration here, as the population of the country is small, the population of those who can work on these Oil fields in 2022 is miniscule. This will increase the diversity of Guyana even more.
Just missing the cut-off is a city like Panama City which has a metro area of 1.5 million which is far too big to qualify as Geneva is just over 1,000,000 in it's metro.

A contender probably not thought about is Miami. The population is around 450K (City of Miami) hosting a huge global banking community, a top 2% presence from all over the world and diplomatic activity from all over the Southern Hemisphere.
Judging by urban or metro population, what would you think the smallest city that has some significance on the global scale is?
Contenders include Geneva (900k metro), Reykjavik, (230k metro) and there are many others as well.
How are you defining "significance on the global scale"? Its global influences, or name recognition? This may not be the same thing.
Bentonville, Arkansas is the headquarters for the largest company in the world in terms of revenue and number of employees and hence plays an outsized role in the global economy for its size. The entire Northwest Arkansas metro area has less than 1 million population. Bentonville itself has around 50,000.
Grand Cayman, Cayman Islands and Hamilton, Bermuda and Nassau, Bahamas are all major offshore banking centers.
Some cities are known for tourism with name recognition but aren't particularly huge including Venice, Nice, Cannes, and Ibiza.

Ibiza (town) on the island of Ibiza only has 50'000 inhabitants.
Lausanne is another contender, being home to the IOC as well as being the smallest city with a metro system and only being 45 minutes from a globally connected airport.
Hell one could consider Pisa a contender due to the leaning tower.
