Civilisations in the Nile and Tigris-Euphrates Valleys seem to have started around the same time, but how long was it before either of them learned of the other's existence? Or don't we know?

People have lived all through that area for literally tens of thousands of years, have been moving about and trading on a small scale for just as long.

I'm sure they knew about each other well before the different cultures evolved into complex civilizations.

They obviously knew about each other but just didn't write it down or didn't write it down on durable writing materials!

Because of the Amarna tablets we know there were very complex diplomatic relations between Egypt and Mesopotamia by at least 3,300+ years ago.

The answer is pre-history. Egypt could not have prospered without Mesopotamian agriculture, and to a lesser extent, animal husbandry.
