I send a generic rejection letter to everyone who applies for a job when I've actually got one posted or have put out word through my network that I'm looking to hire.
I send a personalized email to anyone I do a phone interview with.
I call anyone who I actually bring in for an interview and follow it with a hard copy letter. (I seldom do an in-person interview with more than 2-3 people for any given job.)

I don't do anything, HR handles the notifications for us. We accept applications only online and only for specific openings. They send rejection letters to all who apply, and an offer letter after the phone call to the selected applicant. Those not eligible for an interview get their letter
right away, those interviewed and not hired get a call first followed up by a letter.
Back when we had our business, I don't think we responded to any person we turned down. Actually, the positions were all "subcontractor" deals, but the nature of our line of business was such that we used certain subcontractors very regularly. I think the rules for hiring somebody as a "subcontractor" and hiring them as an actual employee are different.
Personally, I like to get a rejection letter for some closure, be it just a generic (in fact, that's what I prefer).
We send rejection letters to people we have been in touch with. If the resume went straight to the trash, however, we never send out a letter.
